FAQ
Using a micro-spectrophotometer, is it possible to take multiple measurements from one sample?
When the sample is left on the stand for too long, evaporation may occur, and during one measurement, air bubbles may be introduced, resulting in inaccurate measurements, so we recommend reloading each measurement.
What samples can the Bio-Prep series biological sample homogenizers handle?
The biological sample homogenizer is equipped with different grinding beads and procedures to homogenize and crush various samples. The samples that can be processed include animal and plant tissues, soil, feces, fungi, etc. Different samples need to choose different procedures, the specific procedures can be obtained by contacting our application engineers.
What is the possible reason for the residual phenomenon of magnetic beads in the Auto-Pure series automatic nucleic acid extraction instrument?
Reason 1. The program does not match Solution: To optimize the instrument program, you can try to extend the magnetic absorption time or modify the magnetic absorption method to modify it. For liquids that are prone to residual magnetic beads, the magnetic attraction method can be changed to reciprocating magnetic attraction or segmented magnetic attraction in the program settings-options. Reason 2. The kits do not match Solution: Install the kit and the magnetic rod cover correctly, and check whether the kit is loose after it is installed. If there is no problem, the consumables used are slightly different in size from the original matching consumables, and zero calibration is required. For specific methods, please contact our after-sales personnel for consultation.
What are the differences between several Auto-Pure series automatic nucleic acid extractors?
At present, the processing volume of Allsheng automatic nucleic acid extraction instrument is mainly 1 mL, 5 mL and 10 mL. According to the maximum processing volume, the application is also different. Among them, Mini, 32A, 96, etc. with a maximum processing volume of 1 mL are mainly used for routine extraction of viruses; 20B with 5 mL and 24, 24D with 10 mL are mainly used for the extraction of large-volume samples such as cfDNA.
What kinds of samples can Auto-Pure series automatic nucleic acid extraction instruments handle?
The samples that can be processed by the Allsheng automatic nucleic acid extraction instrument include: animal and plant tissues, whole blood, serum, body fluids, swabs, FFPE, etc. Most of the liquid samples such as whole blood and body fluids can be directly extracted and purified for nucleic acid, animals and plants. For solid samples such as tissue and FFPE, the samples should be crushed in advance, or the supernatant should be extracted after high temperature pretreatment outside the instrument.
What are the kits for Auto-Pure series automatic nucleic acid extraction instrument?
Allsheng automatic nucleic acid extraction instrument adopts open software design, and most of the magnetic bead extraction kits on the market can be used on the instrument. If you need to assist in editing the extraction program, you can contact our application engineers.
Auto-Pure series nucleic acid extraction and purification principle?
Auto-Pure series automatic nucleic acid extraction instrument mainly uses magnetic bead method to extract nucleic acid, in which the core of magnetic bead usually has magnetism to realize the transfer function of magnetic rod to magnetic beads. The surface of the magnetic beads is coated with special functional groups to realize the adsorption of nucleic acids or specific substances.
Detection of nucleic acid concentration by ultraviolet absorption method
One of the applications of full wavelength microplate reader Working principle of ultraviolet absorption method: The bases of nucleic acid molecules have the characteristics of absorbing ultraviolet ray, and the maximum absorption wavelength is 260 nm. Under the wavelength of 260 nm, the optical density of 10D value is equivalent to the concentration of double stranded DNA of 50 μg/ml, and it can be used to calculate the concentration of nucleic acid samples. The ordinary microplate reader does not have this function, because it does not have 260 nm, 280 nm, 320 nm light sources and related filters and full wavelength microplate reader (200 nm~850 nm) has this function. However, when using the full wavelength microplate reader, the material also has corresponding requirements: 1.Microplate: UV plate or quartz plate that can pass through ultraviolet light. Ordinary microplate cannot be used! 2.Cuvette: Standard cuvette Micro cuvette Ultramicro cuvette Detection wavelength: 260 nm, 280 nm and 320 nm Results: dsDNA concentration = (A260-A320)x50 (Take dsDAND as an example) dsDNA purity = (A260-A320)/ (A280-A320) Note: our existing AMR-100 microplate reader does not contain this function.
